| Directions | 1. Place a drop of Lactophenol Picric Acid reagent on a clean and dry slide and place fungal culture in it. The stain imparts a yellow colouration on hyphae.
2. By using a nichrome inoculating wire, carefully tease the fungal culture into a thin preparation. 3. Place a cover slip on the preparation. Wait for about 5 minutes and observe first under microscope with low power for screening in low intensity. |

| Principle And Interpretation | Fungus are eukaryotic organism and they are classified into two main groups that is yeast and molds. Its cell wall is made up of chitin. Fungal cells have both macroscopic as well as microscopic structure. Lactophenol Picric Acid is used for staining as well as for wet mounting of fungi. Wet mount preparation is the most widely used method of staining and observing fungi and is simple to prepare. The preparation has three components: Lactic acid preserves the fungal structure and clears the tissue while phenol acts as a disinfectant. Picric acid has antibacterial as well as antifungal properties. It stains the fungal spores and hyphae a pale to dark yellow color. | |||

IF INHALED: Remove to fresh air and keep at rest in a position comfortable for breathing. Absorb spillage to prevent material damage. If on skin or hair: remove/take off immediately all contaminated clothing. Rinse with water/shower. If exposed or concerned: Get medical attention advice. IF SWALLOWED: Immediately call a POISON CENTER or doctor/physician. IF SWALLOWED: Rinse mouth. Do NOT induce vomiting. Specific treatment: refer to Label or MSDS. |
